Considerable hope surrounds the use of disease‐specific pluripotent stem cells to generate models of human disease allowing exploration of pathological mechanisms and search for new treatments. Disease‐specific human embryonic stem cells were the first to provide a useful source for studying certain disease states. The recent demonstration that human somatic cells, derived from readily accessible tissue such as skin or blood, can be converted to embryonic‐like induced pluripotent stem cells (hiPSCs) has opened new perspectives for modelling and understanding a larger (...) number of human pathologies. In this review, we examine the opportunities and challenges for the use of disease‐specific pluripotent stem cells in disease modelling and drug screening. Progress in these areas will substantially accelerate effective application of disease‐specific human pluripotent stem cells for drug screening. (shrink)

Une analyse détaillée d’Aristote, Physique II, 193a10‑27, et de ses commentateurs anciens, ainsi que du Peri Aletheias (F44a Pendrick, col. II, 23‑30) sert de point de départ à une mise à l’épreuve ultérieure de la thèse par (...) la lecture suivie de l’ensemble. Il s’agit de soutenir qu’Antiphon affirme et montre la superposition nécessaire du nomos sur la physis en établissant leur inégal statut ontologique, de substrat matériel d’une part, de forme accidentelle surajoutée d’autre part. Cette interprétation trouve son prolongement dans l’analyse des deux xympheronta (celui de la nature et celui de la norme), et de l’heautos qui leur est couramment associé comme le « soi-même » le serait à « l’utile », selon l’idée qu’il serait question de combattre la norme au nom de l’intérêt personnel supposé, quant à lui, naturel. Le xympheron est plutôt pris ici comme « ce qui constitue », en anthropos, si c’est selon la nature ; en membre d’une communauté politique, si c’est selon la norme. La dépendance mutuelle de la physis et du nomos, comme de leurs xympheronta respectifs, fait alors que le naturel ne se révèle qu’à l’occasion de circonstances accidentelles et éphémères qui, un moment, isolent un membre d’un groupe politique de ce groupe-là, ou au prix d’une opération intellectuelle qui, pour un temps également, fait abstraction de l’œuvre constituante du nomos. Les notions d’équité, de bienveillance à l’égard des êtres humains, de mesure, de pitié interviennent bien dans leurs jugements, mais finissent toujours par se mouler dans le cadre de pensée dominant de la justice rétributive. Nous nous sommes proposés ici de montrer qu’Aristote caractérise le fou (μαινόμενος) dans le cadre d’un système différencié d’autres notions, sans en faire seulement un cas limite, quasiment impensé. Le point de départ de l’étude est l’analyse de la triade ἀκούσιον/δι᾽ ἄγνοιαν/ἀγνοῶν qui convoque aux côtés du fou : l’homme en colère, l’homme pris de vin, celui qui dort, le méchant (μοχθηρός), l’intempérant/incontinent (ἀκρατής), le malade. Cela implique de déterminer les types d’ignorance en cause dans les actes accomplis dans chaque (...) cas et, a contrario, en contraste avec la position socratique, les différentes formes de «l’être dans le savoir» : le savoir en entéléchie première et le savoir en acte ; le savoir du général et celui du particulier ; le savoir en acte sans intervention des affections (πάθη) et le savoir avec affections. Dans l’ensemble des catégories d’acteurs envisagées qui, selon les cas ne mettent pas en acte leur savoir de la différence entre le bien et le mal temporairement, ou le font durablement au point de transformer leur ignorance un état stable (ἕξις) de méchanceté ou encore par choix, comme l’acratique, dans cet ensemble donc, le fou apparaît comme celui qui agit par ignorance (δι᾽ ἄγνοιαν) et non pas de l’un ou de l’autre des paramètres d’une action particulière, mais de tous, à l’exception d’un seul : l’agent car, dit Aristote, «comment s’ignorer soi-même» (ἑαυτόν). Un autre moment de l’article consiste à analyser ce qu’est le soi-même en question et conclut à la réduction de l’αὐτός à ce qui accompagne la mise en acte de l’existence (τὸ εἶναι) que ce soit dans l’agir (πραττειν), le faire (ποιεῖν) ou le vivre (ζῆν). Le fou, en agissant, activerait simplement son sentiment d’exister et en cela ne pourrait «s’ignorer lui-même». Mais il faudra qu’il sorte de cette position et que, revenu à la connaissance de tous les paramètres d’un acte, il se repente de ce qu’il a accompli, pour avoir droit à la qualification d’ἀκούσιος («qui a agi contre son gré»). Chez Aristote la folie semble donc ne pouvoir être philosophiquement prise en compte que comme une crise dont on sort pour mettre en acte son savoir des paramètres d’une action particulière et n’est pas excusable parce que le fou «n’aurait pas eu conscience de son acte» mais parce qu’il a retrouvé la connaissance de ce qui compose un acte. After a quick overview on the specification of the causal role of the powers or faculties (δυνάμεις) in nutrition, from the Hippocratic physicians to Galen, via Aristotle, the article defines the set of questions about this process that the Hippocratic physicians leave open. It then examines how Aristotle provides some sort of indirect physical and ethical answers, while Galen openly takes the side of reappropriating the central Stoic concept of oikeiôsis in a Platonist way.

In this paper, we provide a general setting under which results of normalization of proof trees such as, for instance, the logicality result in equational reasoning and the cut-elimination property in sequent or natural deduction calculi, can be unified and generalized. This is achieved by giving simple conditions which are sufficient to ensure that such normalization results hold, and which can be automatically checked since they are syntactical. These conditions are based on basic properties of elementary combinations of inference rules (...) which ensure that the induced global proof tree transformation processes do terminate. (shrink)

While Peace through Commerce (PTC) started as a conversation among a small group of scholars it has grown into an increasingly robust movement, giving rise to conferences, books, journal articles, and dialogue between scholars, managers, practitioners, government officials, and civil society actors, all of whom share an interest in the potential of commerce to foster greater peace. Because social movement scholarship explores the ability of collective interests to achieve social change it provides a useful lens through which to consider PTC's (...) maturation and, more broadly, the rise of scholarly conversations into social movements its. Increasingly, social movement theory has been used to describe and better understand a diverse range of social and organizational changes (Strang and II Jung, 2005) including academic endeavors (Hambrick and Chen, 2008). I draw on social movement theory to characterize the rise of PTC and to consider its future growth. I also suggest broader implications for the transition of academic dialogue as it attains movement status. (shrink)

In this interview, Cynthia Hammond sits down with Marc Lafrance in order to discuss the 30-year sketching practice that led to her exhibition, Drawings for a Thicker Skin, in 2012. In this practice, Hammond made small, quick drawings of the clothes she would need for trips or key professional events. As she explains, the drawings were not just essential to knowing what to pack; they were essential to being able to pack. While she never conceived of the practice as (...) art, when invited to exhibit the drawings she found a way to relate this idiosyncratic and private practice to a larger set of ontological concerns. The question of whether biologists should continue to use the Linnaean hierarchy has been a hotly debated issue. Invented before the introduction of evolutionary theory, Linnaeus's system of classifying organisms is based on outdated theoretical assumptions, and is thought to be unable to provide accurate biological classifications. Marc Ereshefsky argues that biologists should abandon the Linnaean system and adopt an alternative that is more in line with evolutionary theory. It is often presumed that the laws of nature have special significance for scientific reasoning. But the laws' distinctive roles have proven notoriously difficult to identify--leading some philosophers to question if they hold such roles at all. This study offers original accounts of the roles that natural laws play in connection with counterfactual conditionals, inductive projections, and scientific explanations, and of what the laws must be in order for them to be capable of playing these roles. Conflicts of interest are rampant in the American medical community. Today it is not uncommon for doctors to refer patients to clinics or labs in which they have a financial interest (40% of physicians in Florida invest in medical centers); for hospitals to offer incentives to physicians who refer patients (a practice that can lead to unnecessary hospitalization); or for drug companies to provide lucrative give-aways to entice doctors to use their "brand name" drugs (which are much more expensive than (...) generic drugs). In Medicine, Money and Morals, Marc A. Rodwin draws on his own experience as a health lawyer--and his research in health ethics, law, and policy--to reveal how financial conflicts of interest can and do negatively affect the quality of patient care. He shows that the problem has become worse over the last century and provides many actual examples of how doctors' decisions are influenced by financial considerations. We learn how two California physicians, for example, resumed referrals to Pasadena General Hospital only after the hospital started paying $70 per patient (their referrals grew from 14 in one month to 82 in the next). As Rodwin writes, incentives such as this can inhibit a doctor from taking action when a hospital fails to provide proper service, and may also lead to the unnecessary hospitalization of patients. We also learn of a Wyeth-Ayerst Labs promotion in which physicians who started patients on INDERAL (a drug for high blood pressure, angina, and migraines) received 1000 mileage points on American Airlines for each patient (studies show that promotions such as this have a direct effect on a doctor's choice of drug). Rodwin reveals why the medical community has failed to regulate conflicts of interest: peer review has little authority, state licensing boards are usually ignorant of abuses, and the AMA code of ethics has historically been recommended rather than required. He examines what can be learned from the way society has coped with the conflicts of interest of other professionals --lawyers, government officials, and businessmen--all of which are held to higher standards of accountability than doctors. And he recommends that efforts be made to prohibit and regulate certain kinds of activity (such as kickbacks and self-referrals), to monitor and regulate conduct, and to provide penalties for improper conduct. Our failure to face physicians' conflicts of interest has distorted the way medicine is practiced, compromised the loyalty of doctors to patients, and harmed society, the integrity of the medical profession, and patients. For those concerned with the quality of health care or medical ethics, Medicine, Money and Morals is a provocative look into the current health care crisis and a powerful prescription for change. It is often thought that consciousness has a qualitative dimension that cannot be tracked by science. Recently, however, some philosophers have argued that this worry stems not from an elusive feature of the mind, but from the special nature of the concepts used to describe conscious states. Marc Champagne draws on the neglected branch of philosophy of signs or semiotics to develop a new take on this strategy. The term “semiotics” was introduced by John Locke in the modern period (...) – its etymology is ancient Greek, and its theoretical underpinnings are medieval. Charles Sanders Peirce made major advances in semiotics, so he can act as a pipeline for these forgotten ideas. Most philosophers know Peirce as the founder of American pragmatism, but few know that he also coined the term “qualia,” which is meant to capture the intrinsic feel of an experience. Since pragmatic verification and qualia are now seen as conflicting commitments, Champagne endeavors to understand how Peirce could (or thought he could) have it both ways. The key, he suggests, is to understand how humans can insert distinctions between features that are always bound. Recent attempts to take qualities seriously have resulted in versions of panpsychism, but Champagne outlines a more plausible way to achieve this. So, while semiotics has until now been the least known branch of philosophy ending in –ics, his book shows how a better understanding of that branch can move one of the liveliest debates in philosophy forward. Efforts to bridge emotion theory with neurobiology can be facilitated by dynamic systems (DS) modeling. DS principles stipulate higher-order wholes emerging from lower-order constituents through bidirectional causal processes cognition relations. I then present a psychological model based on this reconceptualization, identifying trigger, self-amplification, and self-stabilization phases of emotion-appraisal states, leading to consolidating traits. The article goes on to describe neural structures and functions involved in appraisal and emotion, as well as DS mechanisms of integration by which they interact. These mechanisms (...) include nested feedback interactions, global effects of neuromodulation, vertical integration, action-monitoring, and synaptic plasticity, and they are modeled in terms of both functional integration and temporal synchronization. To what extent do moral judgments depend on conscious reasoning from explicitly understood principles? We address this question by investigating one particular moral principle, the principle of the double effect. Using web-based technology, we collected a large data set on individuals' responses to a series of moral dilemmas, asking when harm to innocent others is permissible. Each moral dilemma presented a choice between action and inaction, both resulting in lives saved and lives lost. Results showed that: (1) patterns of moral (...) judgments were consistent with the principle of double effect and showed little variation across differences in gender, age, educational level, ethnicity, religion or national affiliation (within the limited range of our sample population) and (2) a majority of subjects failed to provide justifications that could account for their judgments. These results indicate that the principle of the double effect may be operative in our moral judgments but not open to conscious introspection. We discuss these results in light of current psychological theories of moral cognition, emphasizing the need to consider the unconscious appraisal system that mentally represents the causal and intentional properties of human action. In a critical engagement with Hegel's philosophy, Adorno develops a dialectic that is no longer tied to the idealistic concept of a speculative identity of subject and object, yet nevertheless aims at a concept of the absolute. So far neither the scope nor the argumentative consistency of this model of dialectics has been taken into account. Marc Nicolas Sommer reconstructs negative (...) dialectics through its engagement with Hegelian dialectics and defends its central claim: that negative dialectics liberates dialectics from its idealistic shackles and in doing so develops a viable concept of the absolute after Auschwitz. This paper takes up the cause of species pluralism. An argument for species pluralism is provided and standard monist objections to pluralism are answered. A new form of species pluralism is developed and shown to be an improvement over previous forms. This paper also offers a general foundation on which to base a pluralistic approach to biological classification.

What is a fair distribution of resources and other goods when individuals are partly responsible for their achievements? This book develops a theory of fairness incorporating a concern for personal responsibility, opportunities and freedom. With a critical perspective, it makes accessible the recent developments in economics and philosophy that define social justice in terms of equal opportunities. Species pluralism gives us reason to doubt the existence of the species category. The problem is not that species concepts are chosen according to our interests or that pluralism and the desire for hierarchical classifications are incompatible. The problem is that the various taxa we call 'species' lack a common unifying feature.

How should we define ‘health’ and ‘disease’? There are three main positions in the literature. Naturalists desire value-free definitions based on scientific theories. Normativists believe that our uses of ‘health’ and ‘disease’ reflect value judgments. Hybrid theorists offer definitions containing both normativist and naturalist elements. This paper discusses the problems with these views and offers an alternative approach to the debate over ‘health’ and ‘disease’. Instead of trying to find the correct definitions of ‘health’ and ‘disease’ we should explicitly talk (...) about the considerations that are central in medical discussions, namely state descriptions and normative claims . This distinction avoids the problems facing the major approaches to defining ‘health’ and ‘disease’, and it more clearly captures what matters in medical discussions. (shrink)

What altered states of consciousness—the dissolution of feelings of time and self—can tell us about the mystery of consciousness. During extraordinary moments of consciousness—shock, meditative states and sudden mystical revelations, out-of-body experiences, or drug intoxication—our senses of time and self are altered; we may even feel time and self dissolving. These experiences have long been ignored by mainstream science, or considered crazy fantasies. Recent research, however, has located the neural underpinnings of these altered states of mind. In this book, neuropsychologist (...) Marc Wittmann shows how experiences that disturb or widen our everyday understanding of the self can help solve the mystery of consciousness. Wittmann explains that the relationship between consciousness of time and consciousness of self is close; in extreme circumstances, the experiences of space and self intensify and weaken together. He considers the emergence of the self in waking life and dreams; how our sense of time is distorted by extreme situations ranging from terror to mystical enlightenment; the experience of the moment; and the loss of time and self in such disorders as depression, schizophrenia, and epilepsy. Dostoyevsky reported godly bliss during epileptic seizures; neurologists are now investigating the phenomenon of the epileptic aura. Wittmann describes new studies of psychedelics that show how the brain builds consciousness of self and time, and discusses pilot programs that use hallucinogens to treat severe depression, anxiety, and addiction. If we want to understand our consciousness, our subjectivity, Wittmann argues, we must not be afraid to break new ground. Hempel and Giere contend that the existence of provisos poses grave difficulties for any regularity account of physical law. However, Hempel and Giere rely upon a mistaken conception of the way in which statements acquire their content. By correcting this mistake, I remove the problem Hempel and Giere identify but reveal a different problem that provisos pose for a regularity account — indeed, for any account of physical law according to which the state of affairs described by a law-statement presupposes (...) a Humean regularity. These considerations suggest a normative analysis of law-statements. On this view, law-statements are not distinguished from accidental generalizations by the kind of Humean regularities they describe because a law-statement need not describe any Humean regularity. Rather, a law-statement says that in certain contexts, one ought to regard the assertion of a given type of claim, if made with justification, as a proper way to justify a claim of a certain other kind. In wildlife conservation work, rewilding - to make wild once again - refers to the creation of corridors between preserved lands that allow declining populations to rebound. Marc Bekoff, one of our most engaging animal experts and activists, here applies the concept to human attitudes. This paper is concerned with the problem of selfidentification in the domain of action. We claim that this problem can arise not just for the self as object, but also for the self as subject in the ascription of agency. We discuss and evaluate some proposals concerning the mechanisms involved in selfidentification and in agencyascription, and their possible impairments in pathological cases. We argue in favor of a simulation hypothesis that claims that actions, whether overt or covert, are centrally simulated (...) by the neural network, and that this simulation provides the basis for action recognition and attribution. Homeostatic Property Cluster (HPC) theory suggests that species and other biological taxa consist of organisms that share certain similarities. HPC theory acknowledges the existence of Darwinian variation within biological taxa. The claim is that “homeostatic mechanisms” acting on the members of such taxa nonetheless ensure a significant cluster of similarities. The HPC theorist’s focus on individual similarities is inadequate to account for stable polymorphism within taxa, and fails properly to capture their historical nature. A better approach is to treat distributions (...) of traits in species populations as irreducible facts, explained in terms of selection pressures, genealogy, and other evolutionary factors. We call this view Population Structure Theory (PST). PST accommodates the view, implicit in biological systematics, that species are identified by reference to particular historical populations. This paper examines the species problem in microbiology and its implications for the species problem more generally. Given the different meanings of ‘species’ in microbiology, the use of ‘species’ in biology is more multifarious and problematic than commonly recognized. So much so, that recent work in microbial systematics casts doubt on the existence of a prokaryote species category in nature. It also casts doubt on the existence of a general species category for all of life (one that includes both prokaryotes (...) and eukaryotes). Prokaryote biology also undermines recent attempts to save the species category, such as the suggestion that species are metapopulation lineages and the idea that ‘species’ is a family resemblance concept. Donald Davidson's work has been of seminal importance in the development of analytic philosophy and his views on the nature of language, mind and action remain the starting point for many of the central debates in the analytic tradition. His ideas, however, are complex, often technical, and interconnected in ways that can make them difficult to understand. This introduction to Davidson's philosophy examines the full range of his writings to provide a clear succinct overview of his ideas. This book begins (...) with an account of the assumptions and structure of Davidson's philosophy of language, introducing his compositionalism, extensionalism and commitment to a Tarski-style theory of truth as the model for theories of meaning. It goes on to show how that philosophical framework is to be applied and how it challenges the traditional picture.
