Cloning and Expression

Communi et al. (1997) screened a human placenta cDNA library with a human P2Y4 (300038) probe. They isolated a novel clone that encoded a 371-amino acid polypeptide. The novel receptor, termed P2Y11, displayed 33% amino acid identity to the human P2Y1 receptor. Northern blot analysis revealed a 2-kb mRNA with strongest signal in spleen, a weaker signal in small intestine, and no signal in other tissues. Functional expression showed that the P2Y11 receptor is coupled to the stimulation of both the phosphoinositide and adenylyl cyclase pathways and behaves as a selective purinoceptor.

PPAN-P2RY11 Spliced Read-Through Transcript

During the screening of a placenta cDNA library to isolate new P2Y receptors, Communi et al. (2001) cloned a cDNA representing a PPAN (607793)-P2RY11 read-through transcript. See 607793 for information on PPAN-P2RY11.


Gene Function

Adrian et al. (2000) analyzed the expression of several purinergic receptors during differentiation in a promyelocytic leukemia cell line. Granulocytic differentiation was induced by dimethylsulfoxide, and a monocytic/macrophage phenotype was induced by phorbol esters. P2Y11 was detected at relatively low levels in undifferentiated cells, and expression was upregulated during differentiation to both granulocytes and monocytic cells.


Gene Structure

Communi et al. (2001) determined that the P2RY11 gene contains 2 exons.


Mapping

Communi et al. (2001) mapped the P2RY11 gene to chromosome 19p31, where it is contiguous with the SSF1 gene. Since chromosome 19p31 is not a valid location, the P2RY11 gene was presumed to be on chromosome 19p13, and this localization was confirmed by genomic sequence analysis.


Molecular Genetics
