Nlg3KO127/Nlg3KO127 and Nlg3KO127/Nlg3KO88 and Nlg3KO127 hemizygous mutant adults show decreased daily locomotor activity, as compared to controls.

The expression of either Nlg3Δ10.UAS or Nlg3UAS.cXa under the control of Scer\GAL4how-24B rescues the increased bouton number in the third instar larval neuromuscular junction of Nlg3KO127, even leading to fewer boutons compared to controls.

Nlg3KO127 homozygous mutant adults do not show any defects in the medulla columnar restriction of the axonal branches of L4 lamina neurons.

Excitatory junctional potential (EJP) amplitude and miniature excitatory junctional potential (mEJP) frequency are significantly reduced at the mutant third larval instar neuromuscular junction compared to wild type. mEJP amplitude is not significantly affected.

Nlg3KO127/Nlg3f00777 third larval instar larvae show a significant increase in bouton number and a significant decrease in bouton size at the neuromuscular junction compared to wild type. Synaptic branch number at the neuromuscular junction is not significantly affected.

The expression of Nlg3UAS.cXa under the control of Scer\GAL4how-24B rescues the increased bouton number in the third instar larval neuromuscular junction of Nlg3KO127 mutants, even leading to fewer boutons compared to controls.

Muscle-specific expression of Nlg3Scer\UAS.cXa (under the control of Scer\GAL4Mhc.PU) rescues the defects in bouton number and size seen at the neuromuscular junction in Nlg3KO88/Nlg3KO127 larvae. Neuron-specific expression (under the control of Scer\GAL4elav.PU) does not rescue this phenotype.
