Macrophage-specific expression of SCARNIG.4636R using the Scer\GAL4Hml.Δ driver completely disrupts lamellipodia formation.
Expression of SCARNIG.4636R in macrophages from larval stage onwards (under the control of Scer\GAL4Hml.Δ) results in an overall reduction of the actin meshwork and exhibit prominent filopodia consisting of densely packed bundles of actin filaments.
Laser-induced single cell ablation or wounding of the wing epithelial cells in wild-type flies results in cells migrating over long distances to the wound site, while in flies expressing SCARNIG.4636R under the control of Scer\GAL4Hml.Δ the cells fail to migrate towards the wound. Knockdown cells send long filopodial protrusions toward the wound, indicating that the chemotactic response is not affected.
Eye disc specific expression of SCARNIG.4636R under the control of Scer\GAL4GMR.PU does not affect axonal targeting in the visual system.
Third instar larvae expressingSCARNIG.4636R under the control of the pan-neuronal drivers Scer\GAL4elav-C155 or Scer\GAL4sca.PU display axonal targeting defects.
