Constitutive expression of Acf1Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4αTub84B.PL is lethal.
Overexpression of Acf1Scer\UAS.T:Zzzz\FLAG under the control of Scer\GAL4ey.PH results in gross abnormalities in the adult eye, ranging from areas that lack ommatidia, the outgrowth of antenna-like structures from the eye field, oversized or deformed eyes and antannal duplication. These phenotypes are highly penetrant regardless of the transgene insertion site, as they are observed 65-77% of the adult flies analysed.
About half of the eye imaginal discs show obvious morphological abnormalities and irregularities in photoreceptor cell arrangement upon expression of Acf1Scer\UAS.T:Zzzz\FLAG with the Scer\GAL4ey.PH driver.
Overexpression of Acf1Scer\UAS.T:Zzzz\FLAG using Scer\GAL4en.PU or Scer\GAL4da.G32 drivers results in normal adult fly structures.
No abnormalities result from the overexpression of Acf1Scer\UAS.T:Zzzz\FLAG in post-mitotic photoreceptor cells posterior to the morphogenetic furrow using the Scer\GAL4GMR.PF driver.
Scer\GAL4ey.PH>Acf1Scer\UAS.T:Zzzz\FLAG-expression leads to a delay asynchrony of S phase onset along the morphogenetic furrow.
The distribution of apoptotic cells in eye discs expressing Scer\GAL4ey.PH>Acf1Scer\UAS.T:Zzzz\FLAG is not profoundly different from that of controls. However, in some Scer\GAL4ey.PH>Acf1Scer\UAS.T:Zzzz\FLAG discs, but never in controls, apoptotic cells are noticed which are aligned parallel to the morphogenetic furrow.
Expression of Acf1Scer\UAS.T:Zzzz\FLAG in salivary gland cells under the control of Scer\GAL42314 probably interferes with heterochromatin formation, because the banding pattern and size of nuclei with polytene chromosomes appear abnormal. Although a certain degree of variability is observed, the nuclei where Acf1Scer\UAS.T:Zzzz\FLAG is expressed appear to be generally larger than in controls.
