This interesting paper confirms the involvement of transient receptor potential vanilloid (TRPV)-1 ion channels in micturition disturbances, and suggests the use of a TRPV1 antagonist (JTS-653) for the treatment of bladder overactivity. The TRPV ion channel family contains six mammalian members: TRPV1 to TRPV6 (see {1} and references therein). The TRPV1 channel is predominantly expressed in small-diameter primary afferent neurones and is a prominent participator in sensory transduction. Mice lacking the TRPV1 channel have altered micturition thresholds, suggesting that TRPV1 channels may play a role in the detection of bladder filling {2}. JTS-653 competitively antagonised the capsaicin-induced activation of TRPV1 and also inhibited proton-induced activation of human and rat TRPV1. Electrophysiological studies indicated that JTS-653 blocked heat-induced inward currents in rat TRPV1 showing weak or no inhibitory effects on other TRP channels, receptors, and enzymes {3}.
